Quality aspects of Internet telephony
Internet telephony has had a tremendous impact on how people communicate.
Many now maintain contact using some form of Internet telephony.
Therefore the motivation for this work has been to address the quality aspects
of real-world Internet telephony for both fixed and wireless telecommunication.
The focus has been on the quality aspects of voice communication,
since poor quality leads often to user dissatisfaction. The scope of the work
has been broad in order to address the main factors within IP-based voice
communication.
The first four chapters of this dissertation constitute the background
material. The first chapter outlines where Internet telephony is deployed
today. It also motivates the topics and techniques used in this research.
The second chapter provides the background on Internet telephony including
signalling, speech coding and voice Internetworking. The third chapter
focuses solely on quality measures for packetised voice systems and finally
the fourth chapter is devoted to the history of voice research.
The appendix of this dissertation constitutes the research contributions.
It includes an examination of the access network, focusing on how calls are
multiplexed in wired and wireless systems. Subsequently in the wireless
case, we consider how to handover calls from 802.11 networks to the cellular
infrastructure. We then consider the Internet backbone where most of our
work is devoted to measurements specifically for Internet telephony. The
applications of these measurements have been estimating telephony arrival
processes, measuring call quality, and quantifying the trend in Internet telephony
quality over several years. We also consider the end systems, since
they are responsible for reconstructing a voice stream given loss and delay
constraints. Finally we estimate voice quality using the ITU proposal PESQ
and the packet loss process.
The main contribution of this work is a systematic examination of Internet
telephony. We describe several methods to enable adaptable solutions
for maintaining consistent voice quality. We have also found that relatively
small technical changes can lead to substantial user quality improvements.
A second contribution of this work is a suite of software tools designed to
ascertain voice quality in IP networks. Some of these tools are in use within
commercial systems today

High-frequency information content in end-user foreign exchange order flows
This article considers the impact of foreign exchange (FX) order flows on contemporaneous and future stock market returns using a new database of customer order flows in the euro-dollar exchange rate market as seen by a leading European bank. We do not find clear contemporaneous relationships between FX order flows and stock market changes at high frequencies, but FX flows do appear to have significant power to forecast stock index returns over 1–30 min horizons, after controlling for lagged exchange rate and stock market returns. The effects of order flows from financial customers on future stock market changes are negative, while the effects of corporate orders are positive. The latter results are consistent with the premise that corporate order flows contain dispersed, passively acquired information about fundamentals. Thus, purchases of the dollar by corporate customers represent good news about the state of the US economy. Importantly, though, there also appears to be extra information in corporate flows which is directly relevant to equity prices over and above the impact derived from stock prices reacting to (predicted) exchange rate changes. Our findings suggest that financial customer flows only affect stock prices through their impact on the value of the dollar
The Chlamydia muridarum plasmid revisited : new insights into growth kinetics.
Background: Research in chlamydial genetics is challenging because of its obligate intracellular developmental cycle. In vivo systems exist that allow studies of different aspects of basic biology of chlamydiae, the murine Chlamydia muridarum model is one of great importance and thus an essential research tool. C. muridarum carries a plasmid that has a role in virulence. Our aim was to compare and contrast the C. muridarum plasmid-free phenotype with that of a chromosomally isogenic plasmid-bearing strain, through the inclusion phase of the developmental cycle. Methods: We measured infectivity for plasmid bearing and plasmid-cured C. muridarum by inclusion forming assays in McCoy cells and in parallel bacterial chromosome replication by quantitative PCR, throughout the developmental cycle. In addition to these studies, we have carefully monitored chlamydial inclusion formation by confocal microscopy and transmission electron microscopy. A new E.coli/chlamydial shuttle vector (pNigg::GFP) was constructed using standard cloning technology and used to transform C. muridarum for further phenotypic studies. Results: We have advanced the definition of the chlamydial phenotype away from the simple static observation of mature inclusions and redefined the C. muridarum plasmid-based phenotype on growth profile and inclusion morphology. Our observations on the growth properties of plasmid-cured C. muridarum challenge the established interpretations, especially with regard to inclusion growth kinetics. Introduction of the shuttle plasmid pNigg::GFP into plasmid-cured C. muridarum restored the wild-type plasmid-bearing phenotype and confirmed that loss of the plasmid was the sole cause for the changes in growth and chromosomal replication. Conclusions: Accurate growth curves and sampling at multiple time points throughout the developmental cycle is necessary to define plasmid phenotypes. There are subtle but important (previously unnoticed) differences in the overall growth profile of plasmid-bearing and plasmid-free C. muridarum. We have proven that the differences described are solely due to the plasmid pNigg
A T8.5 Brown Dwarf Member of the Xi Ursae Majoris System
The Wide-field Infrared Survey Explorer has revealed a T8.5 brown dwarf (WISE
J111838.70+312537.9) that exhibits common proper motion with a
solar-neighborhood (8 pc) quadruple star system - Xi Ursae Majoris. The angular
separation is 8.5 arc-min, and the projected physical separation is about 4000
AU. The sub-solar metallicity and low chromospheric activity of Xi UMa A argue
that the system has an age of at least 2 Gyr. The infrared luminosity and color
of the brown dwarf suggests the mass of this companion ranges between 14 and 38

Hard X-ray Emission Associated with White Dwarfs
We have used the WGACAT to search for hard X-ray sources associated with
white dwarfs (WDs) from the catalog of McCook & Sion (1999). We find 17 X-ray
sources coincident with WDs showing significant hard X-ray emission at energies
>0.5 keV. Twelve of these WDs are in known binary systems, in two of which the
accretion of the close companion's material onto the white dwarf produces the
hard X-ray emission, and in the other ten of which the late-type companions'
coronal activity emits hard X-rays. One WD is projected near an AGN which is
responsible for the hard X-ray emission. The remaining four WDs and two
additional white dwarfs with hard X-ray emission appear single. The lack of
near-IR excess from the apparently single WDs suggests that either X-ray
observations are more effective than near-IR photometry in diagnosing faint
companions or a different emission mechanism is needed. It is intriguing that
50% of the six apparently single WDs with hard X-ray emission are among the
hottest WDs. We have compared X-ray properties of 11 hot WDs with different
spectral types, and conclude that stellar pulsation and fast stellar winds are
not likely the origin of the hard X-ray emission, but a leakage of the
high-energy Wien tail of emission from deep in the stellar atmosphere remains a
tantalizing source of hard X-ray emission from hot DO and DQZO WDs. (This

W(h)ither the academy? An exploration of the role of university social work in shaping the future of social work in Europe
A controversial proposal to pilot the training of child protection social workers through an intensive work-based route in England is being supported and funded by the UK Government. Frontline, the brainchild of a former teacher, locates social work training within local authorities (‘the agency’) rather than university social work departments (‘the academy’) and has stimulated debate amongst social work academics about their role in shaping the direction of the profession. As a contribution to this debate, this paper explores the duality of social work education, which derives its knowledge from both the academic social sciences and the experience of practice within social work agencies. While social work education has traditionally been delivered by the academy, this paper also explores whether the delivery of training in the allied professions of probation and nursing by ‘the agency’ is equally effective. Finally, this paper explores the Helsinki model which achieves a synergy of ‘academy’ and ‘agency’. It suggests that there are alternative models of social work education, practice and research which avoid dichotomies between the ‘academy’ and the ‘agency’ and enable the profession to be shaped by both social work academics and practitioners
A Study of the Diverse T Dwarf Population Revealed by WISE
We report the discovery of 87 new T dwarfs uncovered with the Wide-field
Infrared Survey Explorer (WISE) and three brown dwarfs with extremely red
near-infrared colors that exhibit characteristics of both L and T dwarfs. Two
of the new T dwarfs are likely binaries with L7+/-1 primaries and mid-type T
secondaries. In addition, our follow-up program has confirmed 10 previously
identified T dwarfs and four photometrically-selected L and T dwarf candidates
in the literature. This sample, along with the previous WISE discoveries,
triples the number of known brown dwarfs with spectral types later than T5.
Using the WISE All-Sky Source Catalog we present updated color-color and
color-type diagrams for all the WISE-discovered T and Y dwarfs. Near-infrared
spectra of the new discoveries are presented, along with spectral
classifications. To accommodate later T dwarfs we have modified the integrated
flux method of determining spectral indices to instead use the median flux.
Furthermore, a newly defined J-narrow index differentiates the early-type Y
dwarfs from late-type T dwarfs based on the J-band continuum slope. The K/J
indices for this expanded sample show that 32% of late-type T dwarfs have
suppressed K-band flux and are blue relative to the spectral standards, while
only 11% are redder than the standards. Comparison of the Y/J and K/J index to
models suggests diverse atmospheric conditions and supports the possible
re-emergence of clouds after the L/T transition. We also discuss peculiar brown
dwarfs and candidates that were found not to be substellar, including two Young
Stellar Objects and two Active Galactic Nuclei. The coolest WISE-discovered
brown dwarfs are the closest of their type and will remain the only sample of

Co-evolution of genomes and plasmids within Chlamydia trachomatis and the emergence in Sweden of a new variant strain.
BACKGROUND: Chlamydia trachomatis is the most common cause of sexually transmitted infections globally and the leading cause of preventable blindness in the developing world. There are two biovariants of C. trachomatis: 'trachoma', causing ocular and genital tract infections, and the invasive 'lymphogranuloma venereum' strains. Recently, a new variant of the genital tract C. trachomatis emerged in Sweden. This variant escaped routine diagnostic tests because it carries a plasmid with a deletion. Failure to detect this strain has meant it has spread rapidly across the country provoking a worldwide alert. In addition to being a key diagnostic target, the plasmid has been linked to chlamydial virulence. Analysis of chlamydial plasmids and their cognate chromosomes was undertaken to provide insights into the evolutionary relationship between chromosome and plasmid. This is essential knowledge if the plasmid is to be continued to be relied on as a key diagnostic marker, and for an understanding of the evolution of Chlamydia trachomatis. RESULTS: The genomes of two new C. trachomatis strains were sequenced, together with plasmids from six C. trachomatis isolates, including the new variant strain from Sweden. The plasmid from the new Swedish variant has a 377 bp deletion in the first predicted coding sequence, abolishing the site used for PCR detection, resulting in negative diagnosis. In addition, the variant plasmid has a 44 bp duplication downstream of the deletion. The region containing the second predicted coding sequence is the most highly conserved region of the plasmids investigated. Phylogenetic analysis of the plasmids and chromosomes are fully congruent. Moreover this analysis also shows that ocular and genital strains diverged from a common C. trachomatis progenitor. CONCLUSION: The evolutionary pathways of the chlamydial genome and plasmid imply that inheritance of the plasmid is tightly linked with its cognate chromosome. These data suggest that the plasmid is not a highly mobile genetic element and does not transfer readily between isolates. Comparative analysis of the plasmid sequences has revealed the most conserved regions that should be used to design future plasmid based nucleic acid amplification tests, to avoid diagnostic failures
Longitudinal dynamics of clonal hematopoiesis identifies gene-specific fitness effects
Clonal hematopoiesis of indeterminate potential (CHIP) increases rapidly in prevalence beyond age 60 and has been associated with increased risk for malignancy, heart disease and ischemic stroke. CHIP is driven by somatic mutations in hematopoietic stem and progenitor cells (HSPCs). Because mutations in HSPCs often drive leukemia, we hypothesized that HSPC fitness substantially contributes to transformation from CHIP to leukemia. HSPC fitness is defined as the proliferative advantage over cells carrying no or only neutral mutations. If mutations in different genes lead to distinct fitness advantages, this could enable patient stratification. We quantified the fitness effects of mutations over 12 years in older age using longitudinal sequencing and developed a filtering method that considers individual mutational context alongside mutation co-occurrence to quantify the growth potential of variants within individuals. We found that gene-specific fitness differences can outweigh inter-individual variation and, therefore, could form the basis for personalized clinical management
